Dr. Goncharova is a member of Dr. Krymskaya’ research team. Dr. Goncharova’s work is focused on defining the role of tumor suppressor proteins, Tuberous Sclerosis Complex 1 (TSC1) and TSC2, in cell growth and motility as they relate to the pathobiology of pulmonary lymphangioleiomyomatosis (LAM). Additionally, her research interests include molecular mechanisms of human airway and vascular smooth muscle cell migration. Dr. Goncharova made the critical observation that TSC2 negatively regulates the activity of the mTOR/S6K1 signaling pathway and found that treatment of LAM cells with rapamycin, a specific inhibitor of mTOR, abrogated LAM cell growth, which identified rapamycin as a promising therapeutic strategy to inhibit cell proliferation in LAM. She also identified TSC1 and TSC2 as modulators of cell migration and invasiveness via regulation of small GTPases Rac and Rho, actin cytoskeleton and focal adhesions, which might be important in metastatic LAM cell growth. Dr. Goncharova’s current research interests are focused on establishing the role of Rho GTPase in modulating cell proliferation in LAM.
